You just want to have the tank mainly empty so when making the change you will have mainly E85 and not a mix of both which would just lower the octane rating of the gas in your car.

Tobz is right, just come with the least amout of pump gas as you can and bring 2 gas cans with you to the tune and fill up there before you get on the dyno. that is the best way and that is what someone else did and it work out for him.
